Pregnenolone Sulfate
Preg-S โข PS โข Pregnenolone-3-sulfate โข Molecule E โข 5-Pregnen-3beta-SO4-20-one
Overview
Pregnenolone Sulfate (PS) is the sulfated form of pregnenolone, and it is pharmacologically opposite to most neurosteroids. While allopregnanolone and THDOC enhance GABAA (calming), pregnenolone sulfate inhibits GABAA and enhances NMDA (excitatory/cognitive).
This makes PS a unique neurosteroid with stimulant-like cognitive effects rather than sedative effects.
In the pheromone community, PS is referred to as "Molecule E" and was discussed in a dedicated PheroTruth thread titled "Molecule E - Pregnenolone Sulfate (5-Pregnen-3beta-SO4-20-one K)." The thread explored its cognitive-enhancing properties and potential synergy with pheromone stacks.
Note on CAS number: CAS 1852-38-6 refers to the sodium salt form (pregnenolone sulfate sodium salt), which is the commercially available form. The free acid form has CAS 1247-64-9 (PubChem CID 105074).
Chemistry
The sulfate group at the 3-position:
- Makes the molecule water-soluble
- Prevents conversion to other steroids
- Creates a negative charge at physiological pH
- Enables unique receptor interactions
PS cannot be converted back to pregnenolone in most tissues (unlike DHEAS to DHEA), so it acts as a distinct compound.
Mechanism of Action
NMDA Receptor Enhancement
PS is a positive allosteric modulator of NMDA receptors:
- Potentiates glutamate responses 5-fold
- Slows receptor deactivation
- Enhances calcium influx
- Increases neuroplasticity
This is the opposite of most neurosteroids (which inhibit NMDA).
GABAA Receptor Inhibition
PS is a negative allosteric modulator of GABAA receptors:
- Reduces GABA-mediated inhibition
- Counteracts sedative effects
- Increases neuronal excitability
- Opposite of allopregnanolone
TRPM3 Activation
PS activates TRPM3 channels:
- Calcium entry
- May affect insulin release
- Sensory modulation
Effects
Cognitive Enhancement
- Memory enhancement -- particularly memory formation
- Improved learning
- Enhanced attention and focus
- Increased neuroplasticity
- Protection against NMDA hypofunction
Excitatory Effects
- Increased alertness
- Reduced sedation
- May increase anxiety at high doses
- Pro-convulsant at very high doses
Pheromone Community Use
PheromoneXS sells pregnenolone (the free base, not the sulfate) as NENO and describes it as a pheromone neurosteroid with the following attributed effects:
- Mood elevation and well-being โ "friendliness and happy feelings"
- Enhanced sensory perception โ "enhancing one's own sense of smell and sight"
- Social signaling โ described as signaling that "the wearer was so healthy and awash in pregnenolone those exposed would have to take notice"
- Cognitive enhancement โ community researchers reported "increased cognitive functions"
PheromoneXS classifies pregnenolone as pheromone-active, though the sulfate form (this page) is the more pharmacologically potent NMDA/GABAA modulator. In the body, pregnenolone and pregnenolone sulfate interconvert, so topical pregnenolone application may produce some of the same neurosteroid effects.
Note: Pregnenolone sulfate itself is too polar and charged to be volatile. Its effects are neurosteroid-mediated self-effects rather than airborne signaling. Valued in stacks for cognitive optimization.
Dosage Guidelines
| Level | Amount |
|---|---|
| Light | 10-25 mg |
| Moderate | 50-100 mg |
| Strong | 150+ mg |
Cautions:
- Can increase anxiety in sensitive individuals
- May lower seizure threshold at high doses
- Opposite effects to calming neurosteroids
- Not for evening use (stimulating)
Comparison to Related Compounds
| Compound | GABAA | NMDA | Net Effect |
|---|---|---|---|
| Allopregnanolone | Enhances | -- | Calming, sedative |
| Pregnenolone | Weakly inhibits | Enhances | Mildly stimulating |
| Pregnenolone Sulfate | Inhibits | Strongly enhances | Stimulating, cognitive |
| THDOC | Enhances | -- | Calming, anxiolytic |
PS is the most excitatory endogenous neurosteroid.
Comparison to Epiallopregnanolone
Both counter GABAA enhancement, but differently:
| Compound | Mechanism | Effect |
|---|---|---|
| Pregnenolone Sulfate | GABAA antagonist + NMDA agonist | Stimulating, cognitive |
| Epiallopregnanolone | Neurosteroid site antagonist only | Normalizing, not stimulating |
PS adds excitation; epiallopregnanolone just removes excess sedation.
Research Applications
PS is studied for:
- Schizophrenia -- NMDA hypofunction model
- Autism -- glutamatergic enhancement
- Cognitive decline -- memory enhancement
- Neuroprotection -- plasticity support
Stacking Considerations
Because PS is excitatory/stimulating:
Good combinations:
- With calming pheromones (buffers them)
- Morning use for cognitive enhancement
- Before mentally demanding tasks
Caution with:
- Anxiety-prone individuals
- Other stimulants
- High androstenone stacks (may increase edginess)
Community Consensus
Rating: 7/10
Valued for cognitive enhancement in the pheromone community. The dedicated PheroTruth "Molecule E" thread demonstrates community interest in its unique NMDA-enhancing properties.
Strengths:
- Real, noticeable cognitive effects
- Unique mechanism (opposite to most neurosteroids)
- Good for focus and memory tasks
- Discussed and tested by community members as "Molecule E"
- PheromoneXS sells the free base (pregnenolone/NENO) as a pheromone molecule with mood/sensory effects
- Used in PheromoneXS blends (Love Boat/XS111, Innocence XS)
Limitations:
- Sulfate form is too polar for airborne signaling โ effects are internal neurosteroid activity
- Can increase anxiety in sensitive individuals
- Stimulating โ not for everyone
- Free base (NENO) vs sulfate distinction is often unclear in community discussions
