Overview
THDOC (3α,5α-tetrahydrodeoxycorticosterone) is an endogenous neurosteroid synthesized from the adrenal hormone deoxycorticosterone (DOC). It's particularly notable as a stress-responsive neurosteroid - levels rise during stress to dampen the HPA axis and reduce anxiety.
THDOC is one of the most potent endogenous positive allosteric modulators of GABAA receptors, particularly those containing the δ-subunit.
Chemistry
THDOC is derived from deoxycorticosterone via two enzymatic steps:
Deoxycorticosterone (DOC)
↓ (5α-reductase type I)
5α-dihydrodeoxycorticosterone
↓ (3α-hydroxysteroid dehydrogenase)

Mechanism of Action
GABAA Modulation
THDOC is a powerful positive allosteric modulator of GABAA receptors:
- Highest potency at δ-subunit containing receptors (>150% enhancement)
- Moderate potency at other isoforms (15-50% enhancement)
- Effects on extrasynaptic (tonic) inhibition
- Synergistic with endogenous GABA
The δ-Subunit Connection
δ-containing GABAA receptors are:
- Located extrasynaptically
- Responsible for tonic (background) inhibition
- Particularly abundant in hippocampus, thalamus, cortex
- Highly sensitive to neurosteroid modulation
This explains THDOC's particularly potent calming effects.
Stress Response Modulation
THDOC plays a key role in HPA axis feedback:
- Stress → increased DOC → increased THDOC
- THDOC potentiates GABA on CRH neurons
- This decreases HPA axis activity

Effects
Alpha THDOC -- "The Reset Pheromone" (HoP #17)
Alpha-THDOC (Allo-THDOC) has been used in mainstream pheromone mixes for only a few years. Its effects include:
- "Hypnotic" spotlight effect: People pay very close attention to the wearer. Produces an invisible "spotlight" that is very attention-grabbing. A trance-like quality when people enter the pheromone cloud.
- Stress and anxiety elimination: Worries and everyday stress get pushed to the backburner. Less "high-strung" behavior. Relaxes without sedating -- you can still function properly.
- Mother-child bonding chemistry: Scientific studies suggest A-THDOC creates a chemical bond similar to that between mother and child. This explains effects like increased sense of closeness between friends, romantic partners, and others exposed.
- Zone-out risk: Very easy to "zone out" while wearing. Can make you concentrate intensely and lose track of time, especially during monotonous tasks. HoP warns this was noticed while driving -- considered unsafe as the wearer focused only on the road directly ahead.
Beta THDOC -- "The Disinhibition Pheromone" (HoP #18)
Beta-THDOC (5b-PREGNAN-3a,21-DIOL-20-ONE, CAS 567-03-3) has distinct effects from the alpha form:
- Disinhibition / truth revelation: Removes the "surface level" face people normally show and reveals their true feelings about you. Can be described as a harsher, more "pushy" version of alpha-THDOC.
- Reduces physical space: If people like you, they move closer. If they dislike you even slightly, they move further away than if you wore nothing at all. Very polarizing.
- Not suitable for new people: People may find it strange that they feel they can talk with complete honesty. Best used with close friends and existing social circles to develop deeper, more meaningful relationships.
- Can be off-putting if used alone: Without other pheromones, people may find you weird or uncomfortable. The forced honesty is too intense without context.
